2012-01-01 The point-spread function (PSF) of a fiber-optic taper-coupled CCD area detector was measured over five decades of intensity using a 20 µm X-ray beam and ∼2000-fold averaging. The ‘tails’ of the PSF clearly revealed that it is neither Gaussian nor Lorentzian, but instead resembles the solid angle subtended by a pixel at a point source of light held a small distance (∼27 µm) above the pixel plane. This converges to an inverse cube law far from the beam impact point. Further analysis revealed that the tails are dominated by the fiber-optic taper, with negligible contribution from the phosphor, suggesting that the PSF of all fiber-coupled CCD-type detectors is best described as a Moffat function. Lin, Wei; Wehrli, Felix W; Song, Hee Kwon 2005-09-01 A technique is proposed for correcting both translational and rotational motion artifacts in magnetic resonance imaging without the need to collect additional navigator data or to perform intensive postprocessing. The method is based on measuring the point spread function (PSF) by attaching one or two point-sized markers to the main imaging object.
Following the isolation of a PSF marker from the acquired image, translational motion could be corrected directly from the modulation transfer function, without the need to determine the object's positions during the scan, although the shifts could be extracted if desired. Rotation is detected by analyzing the relative displacements of two such markers.
The technique was evaluated with simulations, phantom and in vivo experiments. Lew, Matthew D.; Lee, Steven F.; Badieirostami, Majid; Moerner, W. 2011-01-01 We describe the corkscrew point spread function (PSF), which can localize objects in three dimensions throughout a 3.2 µm depth of field with nanometer precision. The corkscrew PSF rotates as a function of the axial (z) position of an emitter. Fisher information calculations show that the corkscrew PSF can achieve nanometer localization precision with limited numbers of photons. We demonstrate three-dimensional super-resolution microscopy with the corkscrew PSF by imaging beads on the surface of a triangular polydimethylsiloxane (PDMS) grating. With 99,000 photons detected, the corkscrew PSF achieves a localization precision of 2.7 nm in x, 2.1 nm in y, and 5.7 nm in z.

Baddeley, David; Carl, Christian; Cremer, Christoph 2006-09-20 To remove the axial sidelobes from 4Pi images, deconvolution forms an integral part of 4Pi microscopy. As a result of its high axial resolution, the 4Pi point spread function (PSF) is particularly susceptible to imperfect optical conditions within the sample. This is typically observed as a shift in the position of the maxima under the PSF envelope.
A significantly varying phase shift renders deconvolution procedures based on a spatially invariant PSF essentially useless. We present a technique for computing the forward transformation in the case of a varying phase at a computational expense of the same order of magnitude as that of the shift invariant case, a method for the estimation of PSF phase from an acquired image, and a deconvolution procedure built on these techniques.
